What Is IGF-1 LR3?
IGF-1 LR3 (Long R3 IGF-1) is a laboratory-modified analog of natural insulin-like growth factor-1 (IGF-1). Natural IGF-1 is a potent growth factor that mediates most of the effects of growth hormone (HGH) and stimulates cell growth and proliferation.
What Does “LR3” Mean?
Two changes in the name radically alter the molecule’s behavior:
- Long — a 13-amino-acid extension has been added to the N-terminus.
- R3 — the amino acid at position 3 has been replaced with arginine.
The combined result of these changes is that the molecule binds much more weakly to IGF-binding proteins (IGFBPs). Whereas natural IGF-1 is “captured” by these proteins in the blood and rapidly neutralized, IGF-1 LR3 stays free. Its half-life is therefore markedly extended and its biological effect is amplified.
Mechanism of Action
- Cell proliferation (hyperplasia) — activates the IGF-1 receptor to stimulate new cell formation and protein synthesis.
- Anabolic signaling — supports growth and repair processes in muscle tissue.
- Glucose metabolism — can lower blood sugar through its insulin-like effect.
Because of these properties, IGF-1 LR3 is used mainly in cell-culture research (as a serum-free medium supplement).
Risks and Limitations
IGF-1 LR3’s potent, non-selective growth stimulus is also its main source of risk:
- It can stimulate growth in all tissues, not only the intended one.
- Its insulin-like effect carries a risk of hypoglycemia.
- Its mitogenic (cell-division-promoting) effect is a serious research question regarding cancer risk.
- Its use for performance or aesthetic purposes in humans is not approved.
